Enhancement of Chitosanase Activity in Bacillus thuringiensis ZJOU-010 Using Response Surface Methodology

Chitooligosaccharides could be produced from chitosan by a newly isolated microorganism, Bacillus thuringiensis ZJOU-010, which exhibited chitosanase activity using shrimp shell wastes as the carbon/nitrogen source. In this study, a 2 central composite experimental design was performed to improve the chitosanase activity by response surface methodology (RSM). The mathematical model developed was adequate and statistically accurate to predict the optimum enzyme activity. The optimum conditions were found to be as follows: shrimp shell powder concentration of 44.98 g/L, (NH4) 2SO4 concentration of 1.48 g/L, and pH of 6.79, under which chitosanase activity of 4.25 U/mL was achieved.
